摘 要:目的通过研究丙烯腈(ACN)对雄性大鼠、小鼠生殖性激素水平的影响,探讨ACN诱导生殖毒性的可能机制。方法选用ICR和昆明种小鼠用不同剂量的ACN(0,6.0,12.0,24.0mg/kg)灌胃染毒8周,选用SD雄性大鼠用不同剂量的ACN(0,7.5,15.0,30.0mg/kg)腹腔注射染毒13周,用放射免疫分析法(RIA)检测血清中睾酮(T)及雌二醇(E2)水平;选用出生后22~25d的SD雌性大鼠,一次性腹腔注射不同剂量的ACN(5.0,15.0,30.0mg/kg)染毒,设阴性对照组(溶剂)和阳性对照组(β-雌二醇,剂量30μg/kg),染毒后6h取子宫称重。结果ACN染毒8周后,ICR小鼠血清中T水平均低于对照组,高剂量组与对照组比较差异有显著性(P<0.01);昆明种小鼠血清中T水平也降低,中剂量组与对照组比较,差异有显著性(P<0.05)。大鼠ACN染毒13周后,低剂量组E2水平明显高于对照组(P<0.05),高剂量组T水平和E2水平明显低于低剂量组(P<0.05),但与对照组比较差异无显著性。雌性SD大鼠染毒后,各剂量组子宫重量与阴性对照组之间差异无显著性,但明显低于阳性对照组。结论ACN能诱导雄性大鼠、小鼠血清性激素水平改变,对雄性大鼠、小鼠生殖内分泌系统有影响,在本实验条件下ACN没有出现雌激素样作用。Objective To study the effects of acrylonitrile(ACN) on sex hormone of rats and mice and to explore the mechanism of ACN on male reproductive toxicity. Methods Male mice were daily given ACN by gavage 5 times a week for 8 weeks at doses of 0,6\^0, 12\^0 or 24\^0 mg/kg body weight; rats were daily administrated ACN ip. 5 times a week for 13 weeks at doses of 0,7\^5,15\^0 or 30\^0 mg/kg body weight, respectively. Levels of testosterone(T)and estradiol(E_2)in sera were measured by radioimmunoassay(RIA). Neonate female SD rats(22~\{25 d\}) were randomly divided into 5 groups: negative control group(solution)and positive control group(β-estradiol,single ip.30 μg/kg),other three groups administrated with ACN single ip.at doses of 5.0,15.0 or 30.0 mg/kg to evaluate whether ACN had estrogen-like activity by uteri-weighing. Results T levels in sera of all treated groups(ICR mice and Kunming mice)were lower than those in control.There was very significant difference in T level between the highest-dose group and control group for ICR mice(P<0.01),and significant difference in T level of sera between the middle-dose group and control group for Kunming mice(P<0.05).E\-2 level was significantly increased in the lowest-dose group after SD rats was treated for 13 weeks(P<0.05).The levels of T and E_2 of sera in the highest-dose group were markedly lower than that in the lowest-dose group,but there was no significant difference between the highest-dose group and control group.There was no significant difference in uteri weight between ACN treated group and negative control group after neonate female SD rats were treated with ACN single ip. Conclusions These results show that ACN could induce changes in sexual hormone levels in sera of male mice and rats.It indicates that ACN may affect reproductive endocrinium of male mice and rats.ACN has no estrogen-like activity.
