摘 要:目的探讨急性排斥反应过程中RANTES基因与蛋白在移植心脏局部表达的意义及环孢素的影响。方法施行大鼠异位心脏移植术,移植大鼠分为3组,每组45只,对照组5只:SD大鼠间的移植为同系移植组(A组),Wistar至SD大鼠的移植分为未用环孢素干预组(B组)及环孢素干预组(C组),健康SD大鼠为对照组。采用RT-PCR方法检测RANTESmRNA的表达,免疫组化方法检测RANTES蛋白的表达。结果RANTESmRNA在A组各时间点和对照组均呈低水平表达,在B组的表达变化与急性排斥反应的进程相关,术后第3天RANTESmRNA表达上调至峰值(3.5±1.27);C组应用环孢素后,RANTESmRNA表达峰值(1.2±0.78)显著低于B组(t=2.18,P<0.05)。RANTES蛋白定位于移植心脏血管内皮细胞与间质单个核浸润细胞。结论RANTES基因与蛋白的表达上调与排斥反应过程中移植物间质单个核细胞浸润密切相关,可能对急性排斥反应的早期诊断有帮助;抑制RANTES信号通路可能是CsA发挥免疫抑制作用的又一分子免疫学机制。Purpose To investigate the expression of RANTES(regulated on activation, normal T cell expressed and secreted) in cardiac allografts and the effect of cyclosporine. Methods Three groups of rats underwent the heterotopic cardiac transplantation,45 cases in each group and 5 cases in control group: SD to SD regarded as isograft group (A group),Wister to SD divided into cyclosporin untreated allograft group (B group) and cyclosporin treated allograft group (C group),normal SD rats as control group.The RANTES mRNA expression was detected by one-step RT-PCR method and the expression of RANTES protein was detected by standard ABC immunohistochemical technique. Results RANTES mRNA was weakly expressed in both A group and control group,and its expression in the B group were correlated with the process of acute allograft rejection.The peak of RANTES mRNA expression 3.5±1.27 in B group appeared in the 3rd day after transplantation,while the peak expression was 1.2±0.78 in C group which appeared in the 7th day,significantly lower than that of B group (t= 2.18, P<0.05).RANTES protein was located in endothelia cells and infiltrating mononuclear cells in allografts?. Conclusions Upregulation of RANTES was significantly correlated with the trafficking of mononuclear cells, which may play an important role in diagnose of acute allograft rejection.It may be one of the important mechanisms of cyclosporin to intervene the acute rejection by inhibit the activation of RANTES pathway.
