摘  要:目的分析大肠杆菌表达的重组人干扰素βSer17(rhIFN-Ser17)的分子结构及其生物学活性。方法通过West-ernblot、氨基酸末端测序、质谱及生物学活性分析等,对大肠杆菌表达的rhIFN-Ser17分子进行鉴定。结果大肠杆菌表达的rhIFN-βSer17的N末端20个氨基酸及C端2个氨基酸序列与文献报道一致,产物的相对分子质量为19877·9,比活性超过2×107IU/mg蛋白。结论大肠杆菌表达的rhIFN-βSer17分子结构与理论推测值完全一致。Objective To analyze the structure and activity of recombinant human interferon β Ser 17 (rhIFN-β Ser 17 ). Methods The rhIFN-β Ser 17 expressed in E.coli was identified by Western blot ,amino acid sequencing at terminals,mass spectrometry and bioactivity assay. Results The sequences of 20 amino acids at N-terminal and 2 amino acids at C-terminal of the expressed rhIFN-β Ser 17 were consistent with those reported.The r elative molecular weight and specific activity of rhIFN-β Ser 17 were 19 877.9 and more than 2×107 IU/mg protein respectively. Conclusion The molecular structure of rhIFN-β Ser 17 expressed in E.coli was com pletely identical to the theoretical value.It laid a foundation of pilot product ion and clinical application of rhIFN-β Ser 17 .
