摘 要:目的研究白细胞介素-2基因佐剂对乙型肝炎疫苗免疫效果的影响。方法利用分子生物学技术构建含IL-2基因的重组质粒pcDNA3.1/IL-2,经测序鉴定、碱裂解法大量提取及Sepharose4FF柱层析纯化,得到重组质粒pcDNA3.1/IL-2和pcDNA3.1/S,免疫豚鼠后,经ELISA检测豚鼠血清中抗HBsAg滴度,CTLL-2细胞株/MTT法检测IL-2活性。结果序列测定和分析结果表明重组质粒pcDNA3.1/IL-2构建正确。血清中抗HBsAg抗体滴度:铝佐剂+HBsAg组为2116mIU;IL-2+HBsAg组为1987mIU;质粒pcDNA3.1/IL-2+HBsAg组为2252mIU;质粒pcDNA3.1+IL-2+pcDNA3.1/S组为67·19mIU;质粒pcDNA3.1/S组为50·88mIU。重组质粒pcDNA3.1/IL-2免疫1周后,IL-2表达水平达高峰,以后逐渐降低,但1个月后IL-2水平仍高于空白对照组。对抗HBsAg抗体滴度和IL-2活性的检测结果进行统计学分析表明,各组间差异有显著意义(P<0·05)。结论IL-2基因佐剂可以增强乙肝重组疫苗和乙肝DNA疫苗的免疫效果。Objective To study the effect of IL-2 gene adjuvant on the immune effect of hepatitis B(H B) vaccine. Methods Construct a recombinant plasmid pcDNA3.1/IL-2 containing IL-2 gene by mole cular biological technique and identify by sequencing.Extract recombinant plasmi d pcDNA3.1/IL-2 and pcDNA3.1/S in a large quantity by alkaline lysis and purify by Sepharose 4FF column chromatography.Divide guinea pigs into different groups and immunize with HBsAg plus different adjuvant including the plasmids.Determin e the anti-HBs titer in the sera of guinea pigs by ELISA,and the IL-2 activity by CTLL-2 cell strain/MTT method. Results Sequencing proved that the recombinant plasmid pcDNA3.1/IL-2 was successfully c onstructed.Twelve weeks after immunization,the anti-HBs titers in the sera of g uinea pigs immunized with aluminium adjuvant+HBsAg,IL-2+HBsAg,pcDNA3.1/IL-2+HB sAg,pcDNA3.1/IL-2+pcDNA3.1/S and pcDNA3.1/S alone were 2 116,1 987,2 252,67.19 and 50.88 mIU respectively.The determination result of IL-2 activity proved t hat the expression level of IL-2 reached a peak value one week after immunizati on with pcDNA3.1/IL-2,then decreased gradually.However,the IL-2 level one mont h after immunization was still higher than that of blank control.Both anti-HBs titers and IL-2 activities were significantly different among the groups. Conclusion IL-2 gene adjuvant enhanced the immune effects of both recombinant HB protein v accine and HB DNA vaccine.
