乳宁冲剂及拆方对裸鼠移植瘤生长及肺转移的影响

摘　　要：通过体内实验探讨乳宁冲剂及其拆方对裸鼠移植瘤生长及肺转移的抑制作用。选雌性BALB/c裸鼠,以对数生长的体外培养的MDA MB435高转移乳腺癌细胞造模。随机分为模型组、乳宁冲剂组、乳宁冲剂拆方温肾组、乳宁冲剂拆方疏肝活血组、三苯氧胺组(TAM组)、环磷酰胺组(CTX组),给药7周后记录移植瘤直径、瘤重,解剖显微镜下计数肺转移结节,病理切片观察肺转移灶大小及流式细胞仪检测瘤组织细胞周期。结果:乳宁冲剂组对肿瘤生长抑制作用明显;乳宁冲剂组及拆方温肾组、理气活血组对裸鼠乳腺癌移植瘤的肺转移抑制作用分别为46.36%、32.73%、18.2%,乳宁冲剂组作用与CTX组(50.91%)比较无显著差异;乳宁冲剂组可抑制细胞的有丝分裂,增加G0/G1期细胞数量,减少S期细胞数目,与模型组比有统计学意义(P<0.05),温肾组也有此作用趋势。乳宁冲剂全方抑制肿瘤生长及抗转移作用优于拆方,而拆方组以温肾组抗转移作用优于疏肝活血组。乳宁冲剂可能通过调整细胞周期,抑制DNA合成而抑制肿瘤生长及转移。To explore the effects of Runing Granule and its detached formula on the restrain growth and anti-metastasis of breast cancer by experimental research in vivo, BALB/c female mice were modeled by transplanting with high metastasis MDA-MB-435 breast tumor cell and were randomly divided into model group,Runing Granule group and its detached formula for regulating thoroughfare-conception vessels and smoothing liver to activate blood groups, tamoxifen (TAM) group, cyclophosphamide(CTX) group. Each group was treated for 7 weeks. The diameter and weight of tumor were recorded by means of anatomical microscope to take count of the nodes of lung surface and lung tissue slice to observe lung metastasis. Facscalibour was adopted to observe the cell cycle. Results: Runing Granule group significantly reduce the growth of tumor. Runing Granule and regulating thoroughfare-conception vessels, or soothing liver to activate blood groups in suppressing lung metastasis were 46.36%, 32.73% and18.2% respectively,there was no significant difference between Runing Granule group and CTX group(50.91%). Runing Granule could inhibit the cell mitosis,increase the ratio of G0/G1, reduce the ratio of S-phase cell. The regulating thoroughfare-conception vessels group had similar effect. The inhibiting tumor growth and anti-metastasis effects of Runing Granule was better than its detached formulae, and the formula for regulating thoroughfare-conception vessels group was better than smoothing liver to activate blood group in suppressing lung metastasis. The anti-metastasis mechanisms of Runing Granule may adjust cell cycle and inhibit the DNA synthesis of tumor.
