摘 要:以霉菌的ITS2基因为靶序列,利用DNAman、DNAstar等生物信息软件在靶序列的指定片段内设计针对黑曲霉、黄曲霉、串珠镰刀菌和鲜绿青霉菌的30bp种特异性寡核苷酸探针和通用引物。将4种标准菌种的基因组通过扩增、线性Cy3或Cy5荧光标记、纯化、杂交试验,验证了探针的特异性和技术的可行性。结果表明,基因芯片可以应用于产毒霉菌的检测。The objective of the research was to study the detection method of four common toxic mold species by genechip technology. The gene-targeted sequences were decided by analyzing DNA sequences of the ITS region. By means of DNAman?DNAstar and blastn in GenBank, the oligonucleotide probes of 30bp were analyzed, designed and selected. Four samples were amplified and lablled with cy3 or cy5 fluorescent, then the purified amplicons were hybridized to the genechip.Based on the results of hybridization experiment, we thought that the detection genechip can be used for detection and identification of common toxic molds.
