军牧1号白猪OB基因cDNA序列分析

摘　　要：采用RT-PCR法获得军牧1号白猪的OB(OBase)基因cDNA,并对其进行了克隆和序列分析.结果发现2种新变异序列:在编码区第364位GCC突变为CGC,导致第122位的Ala突变为Arg;第153～155位的碱基(CAG)发生缺失,使第49位的谷氨酸(Gln)发生缺失.The nucleotide sequence of OB gene complementary deoxyribonucleic acid of Junmu No.1 white pig have been cloned and analyzed by RT-PCR.Two new different mutants genotype on ob gene were found:One mutated at No.364 in the coding sequence of OB gene,causing a mutation at No.122 Ala in the amino acid of OB gene.The other deleted three bases at No.153 to 155 base in the coding sequence of OB gene,leading to the deletion of No.49 Gln in the amino acid sequence of OB gene.The similar reports have found in Japanese and 30% wild type C57BL/6J mouse besides this text reported,in which have found deletion of No.49 Gln codon.
