维生素E对牛核移植供体细胞凋亡率的影响

摘  要:采用光镜观察、DNA琼脂糖凝胶电泳、Hoechst33342荧光染色及流式细胞仪分析等方法,检测了不同浓度的维生素E对新生牛耳缘成纤维细胞凋亡的影响。结果表明,不同浓度的维生素E均可显著降低牛耳缘成纤维细胞的凋亡率,与对照组相比差异极显著(P<0.01),以100μmol/L为最佳浓度。DNA琼脂糖凝胶电泳检测,100、150μmol/L维生素E处理组电泳结果未见梯状条带,对照组和50μmol/L维生素E处理组则出现典型的梯状条带。The effects of vitamin E at different levels of concentration on new born bovine ear fibroblasts apoptosis were detected by optical microscopy,agarose gel electrophoresis.Hoechst 33 342 fluorescence staining and flow cytometry.The result showed that the serum starvation made the apoptosis rate rised significantly as compare to the control,the vitamin E reduced the apoplosis significantly(P<0.01).Thus the optimum inhibitor concentration of vitamin E was 100 μmol/L.The morphological changes of apoptosis were clearly. Under the optical microscope,the characteristics of apoptosis,such as the chromatin condensed and marginated,and apoptotic bodies were observed in fibroblasts.DNA ladders appeared in control and 50 μmol/L treatment groups,but did not appear in 100 μmol/L and 150 μmol/L vitamin E treatment groups.
