摘  要:目的考察淫羊藿消渴冲剂(epimediumhypoglycemicgranules,EHG)对实验性2型糖尿病模型大鼠的作用,并探讨其降糖作用机制。方法用小剂量链脲霉素(30mg·kg-1,静脉注射)加高热量饲料建立实验性2型糖尿病模型大鼠,然后用EHG分3个剂量组(25、50、100mg·kg-1),灌胃给药,治疗4周,并测定空腹血糖(fastingbloodglucose,FBG)、糖耐量、血清胰岛素以及肝细胞膜的胰岛素受体、血脂和肝脏过氧化脂质(LPO)的含量。结果EHG能明显降低实验性2型糖尿病大鼠的FBG,改善其糖耐量异常、降低血脂及肝脏LPO的含量,并使胰岛素水平恢复正常,且呈现一定的量效关系,此外放射配基受体结合实验显示EHG能明显增加肝细胞膜胰岛素受体数目,但对亲和力无明显影响。结论EHG对实验性2型糖尿病大鼠模型有明显的治疗作用,其降糖作用机制与增加肝细胞膜胰岛素受体数目及降低肝脏LPO有关。Objective To study the effect of EHG on type 2 diabetes(T2D)rats and its mechanism.Methods T2D rat was established by low dosage streptozotocin(STZ (30 mg·kg^(-1)),iv)and hyperotophic foods,then treated with EHG(25,50,(100 mg·kg^(-1)),ig) for four weeks.Fasting blood glucose(FBG),serum insulin level,glucose tolerance test,serum fat content and ()^(125)I-insulin receptor to liver plasmamembrane and LPO in liver of the T2D rats were measured.Results EHG could significantly lower FBG,improve glucose tolerance and dyslipidemia and decrease liver LPO content,and make insulin level normal.Moreover,it could also remarkably raise the level of the low-affinity insulin receptor but had no effect on the insulin receptor affinity.Conclusion EHG has an obvious therapeatic effect on T2D rats.Its mechanism is related to improve the insulin receptor number and decrease liver LPO content.
