摘 要:目的通过研究益气活血药对血管紧张素Ⅱ(AngⅡ)致心脏成纤维细胞增殖的影响,寻找能抑制血管紧张素Ⅱ所致心脏成纤维细胞增殖的益气活血中药。方法通过在培养乳鼠心脏成纤维细胞增殖加入血管紧张素Ⅱ,观察细胞直径和细胞数;在此基础上,进行益气活血中药对血管紧张素Ⅱ致培养乳鼠心脏成纤维细胞增殖实验药理学的研究。结果Losartan、丹参素和川芎嗪大、中、小剂量组可显著抑制AngⅡ增加心肌成纤维细胞数量的作用,对心肌成纤维细胞直径无明显影响,说明Losartan、丹参素和川芎嗪抑制心肌成纤维细胞的增殖,且呈一定的量效关系,而益气药党参和黄芪无此作用。结论活血药丹参素、川芎嗪可抑制AngⅡ引起的心脏成纤维细胞增殖并呈一定的量效关系;益气药党参、黄芪在本研究中未发现有此抑制作用。Objective The objective is to search for some Chinese herbs with Qi-tonicity and promoting blood circulation to the proliferation of the myocardial fibroblasts induced by Angiotensin Ⅱ (Ang Ⅱ).Methods The proliferation of the myocardial fibroblasts induced by AngⅡ was analyzed by calculation of the cell diameter and the number of the cell in the cultured neonatal myocardial fibroblasts in vitro.The effects of Chinese herbs on the myocardial fibroblasts induced by angiotensinⅡin the cultured neonatal cardiac cells in vitro were observed. The efficient components of Danshensu, Ligustrazine, and Astrgalus Root injection were studied.The effects of Danshen and Danshen with Dangshensu were analyzed by serum pharmacology.Results AngⅡwith different concentrations of Lgustrazine (large, middle, small). There were no significant differences of the cell diameter for them. The results showed that Danshensu and Lgustrazine as the Losartan could be able to reduce the number of the cells and to prevent the proliferation of myocardial fibroblast. There was the dose-effect relationship in cultured neonatal myocardial fibroblast in vitro for them.There were no the effects for Dangshen,Astrgalus Root,and Dangshen with Danshen(middle and small).Conclusion The results suggest that Chinese herbs with Qi-tonicity and promoting blood circulation could be able to inhibit the proliferation of the myocardial fibroblasts induced by angⅡin the cultured neonatal cardiac fibroblasts in vitro. Based on Chinese traditional theory, there may be that the blood stasis is the e basis of left ventricle hypertrophy (LVH) and to promote blood circulation may be able to prevent LVH.
