摘 要:以华北落叶松(Larixprincipis-rupprechtii)幼苗为研究材料,采用冷驯化和非冷驯化两个处理分别提取RNA,然后逆转录成cDNA,PCR扩增,6%变性丙烯酰胺测序胶电泳,银染显带。结果表明:华北落叶松冷驯化与非冷驯化基因表达有一定差异,经二次扩增初步确定冷驯化处理的扩增产物中在500bp处有一条抗冻特异带。同时,对冷驯化不表达片段进行了分析。Larix principis-rupprechtii seedlings were treated with cold acclimation and nonacclimtion to extract RNA. After treatment, they were reversed transcription as cDNA, PCR amplificated, 6 percent PAGE, and silver stained. The results showed that differential gene expression occurred between cold acclimtion and nonacclimtion of Larix principis-rupprechtii. They were appraised with double amplification after recycling. One antifreeze band was detected primarily and it was 500 bp. Nonexpression gene fragment of larch during cold acclimtion was also analyzed.
