摘  要:目的:观察Ⅱ型糖尿病大鼠血脂的变化、视网膜毛细血管病变及环烯醚萜总苷对其影响。方法:自制高脂饲料喂养Wistar大鼠4周后,按25mg/kg一次腹腔注射链脲佐菌素(streptozotocin,STZ),2周后用口服糖耐量试验(Oralglucosetolerancetest,OGTT)筛选造模成功大鼠。随机分为糖尿病模型组(Model)、降糖通脉胶囊组(JiangtangTongmaiJiaonang,JTTM)及环烯醚萜总苷组(IridoidglycosidesofCornusofficinalis,IGCO),并设正常对照组(Control)。饲养16周,观测血清总胆固醇(CHOL)、甘油三脂(TG)、载脂蛋白A(ApoA)及B(ApoB)、高密度脂蛋白(HDLC)及低密度脂蛋白(LDLC),并运用视网膜消化铺片和图像分析方法,观测糖尿病大鼠视网膜微血管结构改变及环烯醚萜总苷对其影响。结果:模型组大鼠血清ApoA水平明显降低,血清CHOL、TG、LDLC、ApoB水平明显升高。环烯醚萜总苷组能显著升高糖尿病大鼠ApoA水平,降低糖尿病大鼠CHOL、TG、LDLC、ApoB水平。模型组视网膜毛细血管内皮细胞(EC)数目明显增加,而周细胞(PC)数目明显减少,导致E/P值上升。环烯醚萜总苷组均能显著降低糖尿病大鼠视网膜毛细血管的E/P值。结论:环烯醚萜总苷对糖尿病血脂改变及视网膜毛细血管病变具有一定的改善作用。AIM: To investigate the lipo-blood changes and the pathological changes in retinal capillaries of rats with diabetes mellitus and the influence of a traditional Chinese medicine compound named 'IGCO'. Methods:Diabetic animal models were induced by enriching-diet for 4 weeks and STZ on Wistar rats, and successful models were screened by OGTT two weeks later. These rats were divided randomly into diabetes model group, JTTM group, IGCO group and normal control group. After 16 weeks, CHOL、 TG、 Apo-A、 Apo-B、 HDL-C、 LDL-C were observed. The retina was observed by trypsin digestion and figure analysis. Results: In model group, Apo-A decreased and CHOL、 TG、 LDL-C、 Apo-B increased. IGCO could significantly increase Apo-A and decrease CHOL、 TG、 LDL-C、 Apo-B. In model group, the endothelial cells increased and pericyte decreased in retinal capillaries, so E/P increased. IGCO could significantly decrease the E/P. Conclusions: IGCO could improve the lipo-blood and the pathological changes in retinal capillaries of rats with diabetes mellitus.
