摘  要:目的观察人参总皂甙(TotalsaponinofPanaxGinseng,TSPG)和西洋参茎叶总皂甙(Panaxquinquefoliumlsaponin,PNS)对贫血小鼠造血恢复的影响,并比较二者作用的异同。方法采用Co60照射注射环磷酰胺、氯霉素复合造模建立贫血小鼠模型,应用全自动血球分析仪和造血祖细胞体外培养技术,检测外周血和三系造血祖细胞集落。结果TSPG与PNS都可以促进贫血小鼠骨髓有核细胞(BMC)和红细胞总数(RBC)、血红蛋白(HB)、血小板(PLT)的回升(P<0.01或P<0.05),;对白细胞总数(WBC),TSPG与PNS显示相反的作用,TSPG有较弱的提升WBC的作用(P>0.05),PNS则抑制WBC的回升(P<0.05)。在造血祖细胞体外给药培养中,TSPG在25~100μg/mL范围内对三系祖细胞增殖都有促进作用,对红系、巨核系最适刺激浓度是25μg/mL,对粒系的最适刺激浓度是50ug/mL。PNS浓度为50~100μg/mL时对红系、巨核系有促进作用,其中巨核系作用冒险(P<0.01);25~50μg/mL时对粒系增殖没有明显影响,100μg/mL时则表现抑制作用(P<0.05)。结论人参总皂甙和西洋参茎叶总皂甙都可以促进贫血小鼠造血功能的恢复,但其作用各有特点。Objective: TO compare the effect of PNS on hematopoiesis in anemic mice with that of TSPG。 Methods: Anemic mice model resulted from myelosuppression by irradiation and cytotoxic chemotherapeutic compounds were randomly divided into three groups: modle control group,TSPG treatment group,PNS treatment group。hematopietic progenitor cells culture in vitro and peripheral blood count were used to detect the numbers of peripheral blood cells and CFU-GM ,CFU-E/ BFU-E and CFU-Meg. Results: Both of TSPG and PNS could significantly promote the rise of RBC,HB(P<0.05)and BMC(P<0.01).TSPG and PNS showed adverse effect on WBC. TSPG could slightly increase the numbers of WBC(P>0.05), while PNS decreased it in anemic mice(P<0.05). Concentration of TSPG between 25-100μg/ml could promote the proliferation of CFU-GM ,CFU-E/ BFU-E and CFU-Meg. The optimal concentration for growth of CFU-E/ BFU-E and CFU-Meg was 25μg/ml and the optimal concentration for growth of CFU-GM 50μg/ml.Concentration of PNS between 25-100μg/ml had an positive influence on erythropoiesis and magakaryopoiesis. Though PNS in low concentration( 25-50μg/ml) showed no obvious effect on the growth of CFU-GM ,high concentration of PNS(100μg/mL )could suppress the growth of CFU-GM. Conclusion: Both of PNS and TSPG can promote hematopoiesis in anemic mice though they act in different way.
