摘  要:建立了高效液相色谱法测定猪血浆中阿莫西林含量的方法。采用Agillent1100HPLC系统,色谱柱为SBC18,流动相为0.05mol/L磷酸二氢钾-0.1mol/L三乙胺-乙腈(800∶120∶7,pH2.5),流速1.0mL/min,柱温30℃,检测波长220nm。猪血浆样品经高氯酸溶液变性处理,取上清液直接进样测定。阿莫西林的色谱峰面积对其浓度的线性关系范围在0.5~20.0μg/mL之间,相关系数为0.9997。阿莫西林低、中、高(0.5,5.0,20.0μg/mL)三种添加浓度猪血浆样品的平均加样回收率分别为100.7%、105.1%、103.3%(n=3),变异系数分别为3.0%、0.1%、3.3%(n=3)。该法可用于猪血浆中阿莫西林含量的测定和相关的药代动力学研究。A simple and accurate HPLC method with UV detector at 220 nm was developed for the determination of amoxicillin in pig plasma. The sample work-up procedures include 20%(V/V) perchloric acid denaturalization of protein and precipitation of protein by freezing centrifuging. The standard curve of amoxicillin was linear in the range of 0.5~20.0 μg/mL (r=0.999 7, n=7). The RSD for repeated detection at 0.5, 5.0 and 20.0 μg/mL in pig plasma spiked with amoxicillin were 3.0%, 0.1% and 3.3%(n=3) and the average spiked recoveries were 100.7%, 105.1% and 103.3% (n=3), respectively. This method offered a rapid, repeatable and accurate procedure and can be used to determinate pharmacokinetic parameters of amoxicillin in pig plasma.
