摘 要:目的:观察慢性束缚应激时大鼠海马BDNF、TrkB、NT3的变化及逍遥散对其影响。方法:用束缚的方法建立了应激模型,用心脏灌流的方法固定海马,切片后用免疫组织化学结合图像分析的方法观察了海马CA1区BDNF、TrkB、NT3变化情况。结果:发现海马中BDNF和NT3的阳性细胞数和积分光密度下降,TrkB阳性细胞数和积分光密度明显上升。逍遥散能不同程度的抑制海马中BDNF和NT3阳性细胞数和积分光密度的下降以及TrkB阳性细胞数和积分光密度的上升。结论:BDNF、TrkB、NT3在慢性应激中发挥了重要作用,逍遥散可能正是通过它们的变化,作用于HPA轴引起机体免疫功能变化的。Objective:Explore the changes of BDNF,TrkB,NT_3 in hippocampus of rats of chronic immobilization stress model and effect of Xiaoyaosan.Method:Rats model of chronic immobilization stress was made by binding rats on the specific bound frame.Rats were instilled with formamint solution through heart.30μm slices were made in cryotome.Method of immunohistochemistry combined with image analysis was applied to observe the change of BDNF,TrkB,NT_3 in hippocampus.Result:Counts of positive cell and points of optical density of BDNF and NT_3 in hippocampus were lower and above-noted result of TrkB was markedly higher.Xiaoyaosan could improved these parameters.Conclusion:BDNF,TrkB and NT_3 play an important role on the chronic immobilization stress,perhaps Xiaoyaosan cause the changes of BDNF,TrkB,NT_3 and induce the change of immunological function through HPA axis.
