摘  要:目的探讨全血中苯丙氨酸和酪氨酸测定时标本采集与保存对测定结果的影响。方法利用高效液相色谱紫外检测法测定41例EDTA·K2抗凝血浆,肝素抗凝全血及EDTA·K2抗凝全血在室温(25±1)℃、4℃保存下0、24、72、120、168h及冰冻1周后Phe和Tyr的浓度。结果肝素与EDTA·K2两种不同抗凝剂抗凝时全血中苯丙氨酸和酪氨酸测定结果比较无统计学意义(P>0.05)。全血中的Phe和Tyr浓度比血浆中的低。冰冻条件下一周内全血中的Phe和Tyr浓度保持稳定。结论标本的保存处理方式直接影响到测定结果的准确性。Objective To investigate the influencing effect of sample collection and storage measures on phenylalanine and tyrosine measurements in whole-blood samples. Methods The concentration of phenylalanine and tyrosine in plasma and whole-blood samples which were storaged at 4℃,25℃ for 0,24,72,120,168 hours or frozen preserved for one week were determined by high-performance liquid chromatography with ultraviolet-detection. Results The levels of phenylalanine and tyrosine in whole-blood were consistent when using EDTA·K_2 or heparin as anticoagulant (P>0.05). The whole-blood phenylalanine and tyrosine were significantly lower than in plasma. Phenylalanine and tyrosine in whole-blood that frozen stored were stable within one week. Conclusion Sample collection and storage method can significantly affect the levels of phenylalanine and tyrosine in whole-blood measured.
