摘  要:目的探讨雌激素对人精子的激活作用及其可能的作用机制。方法以17β-雌二醇(E2)及非透膜性大分子雌二醇-牛血清白蛋白交联物(E2-BSA)分别作用于生育力正常男性的精子,以三色法染色评价精子顶体反应(AR)发生率,流式细胞术检测精子内游离Ca2+浓度[Ca2+]i的变化。结果E2可引起获能精子AR率明显增加,使[Ca2+]i快速升高,对非获能精子则无明显影响;E2-BSA同样可以提高获能精子AR率及[Ca2+]i,其作用与E2相似;E2诱导获能精子[Ca2+]i升高依赖于胞外Ca2+的内流。结论雌激素对人精子有一定的激活作用,该作用可能是通过与人精子膜上的雌激素结合位点作用后使胞外Ca2+内流而实现的。Objective To study the effects of 17β-estradiol(E_2) on the activation of human spermatozoa and the possible mechanism. Methods Spermatozoa of fertile men were stimulated by E_2 and the membrane impermeable conjugated 17β-estradiol (E_2-BSA) respectively. Then the acrosme reaction (AR) rate was determined by triple-stain technique and the intracellular calcium concentration ([Ca^(2+)]_i) in spermatozoa was measured by flow cytometry. The changes of [Ca^(2+) ]_i were measured again following the addition of E_2 and E_2-BSA in the calcium-free medium. Results E_2 significantly increased the AR rate and the [Ca^(2+)]_i in capacitated human spermatozoa, while it had no effects on the uncapacitated spermatozoa. E_2-BSA also promoted AR and increased [Ca^(2+)]_i in the capacitated human spermatozoa. Additionally, the change of [Ca^(2+)]_i was absent in case that human spermatozoa were stimulated by E_2 or E_2-BSA in the calcium-free medium. Conclusion Estrogen can activate human spermatozoa and such activation is probably mediated by the integrating of estrogen with the specific sites on human spermatozoa and thus causing the influx of extracellular calcium.
