摘 要:目的:观察自体微小颗粒骨对骨髓基质细胞(MSCs)的诱导作用及复合MSCs移植修复大鼠骨缺损的治疗效果.方法:全骨髓培养法获得大鼠MSCs,分别以块状骨和微小颗粒骨诱导MSCs,半定量RTPCR,免疫组化测定细胞碱性磷酸酶(ALP),骨钙素(OCN)和Ι型胶原的表达.制作大鼠颅骨缺损动物模型,培养同种异体新生大鼠的MSCs,复合自体颗粒骨后植入骨缺损区,X线摄片观察骨愈合情况;5溴脱氧尿嘧啶(BRDU)标记观察移植细胞成活情况.结果:MSCs与颗粒骨混合培养,ALP,OCN和Ι型胶原的表达高峰提前,表达水平高于块状骨.植入微小颗粒骨复合MSCs组颅骨缺损愈合优于单纯植入微小颗粒骨,可见BRDU标记细胞存活.结论:微小颗粒骨具有骨诱导活性,复合MSCs移植修复颅骨缺损,缺损愈合加快,移植细胞可存活.AIM: To investigate the induction effect of morselized bone on marrow stromal cells and to study the therapeutic effects of compound autogenetic minimal morselized bone and marrow stromal cells on rat cranial defect. METHODS: Rat marrow stromal cells (MSCs) were cultured and induced with morselized bone and mass bone. ALP, OCN, collagen Ι expressions were semiquantified by RT-PCR and immunohistochemical staining. The rat cranial defect model was prepared. The defect was repaired with compound autogenetic minimal morselized bone graft and marrow stromal cells. X ray examination was performed to observe the union. The survival of grafted cells was examined with BRDU immunohistochemical staining. RESULTS: After cultured with morselized bone, MSCs changed to osteoblast and the expressions of ALP, OCN and collagen Ι in MSCs were significantly earlier and higher than those in mass bone group. The healing of cranial defect was more satisfactory in compound morselized bone and morrow stromal cells grafting group than that in morselized bone group and bromodeoxyuridine (BRDU) positive osteocytes were seen. CONCLUSION: After compound autogenetic minimal morselized bone and marrow stromal cells grafting, rat cranial defect repairs rapidly and the grafted cells survive.
