摘  要:就中药提取物对肿瘤坏死因子(TNF)的诱生作用做了一定的探索和研究。建立了用L-929细胞—结晶紫染色法结合S-180细胞~3HTdR同位素掺入法检测TNF活性,并以此为筛选指标建立了药物筛选模型。用此筛选模型对36味中草药进行了初步筛选发现:分属7个科的7味中草药的乙醇提取物对TNF有不同程度的诱生作用,其中以毛茛科植物猫爪草效果最好。将猫爪草的乙醇萃取液浓缩后,经溶媒分级分离萃取及柱层析分离,得到单一的有效成份,结构分析证明为软脂酸。诱生出的TNF通过单克隆抗体中和试验得到确证。The authors set up a screening system for the detection of TNF inducers. By using this model 36 traditional Chinese medical herbs from 28 families were studied. Seven herbs from 7 families were found to be active in the TNF induction. Among them Ranunculus Ternatus (RT) is the best one. The active components of RT were isolated from the crude extract by solvent extraction and column chromatography.Eleven fractions were obtained. RT-A2,one of the active components, is a white crystal and its IR spectrum is similar to Standard Infrared Grating Spectra No. 28344K. Therefore, RT-A2 may be identified as a fatty acid (palmitic acid). Its optimum dose of inducing TNF is 50μg/ml in vitro. TNF induced in the cell culture was demonstrated by monoclone antibody neutralizing test.
