摘  要:目的研究机械牵张对大鼠心肌组织低氧诱导分子1α(hypoxiainduciblefactor1α,HIF1α)和血管内皮细胞生长因子(vascularendothelialgrowthfactor,VEGF)表达的影响。方法采用大鼠离体灌流心脏模型,膨胀左心室30min,用RTPCR方法观测左室心肌细胞HIF1α、VEGF的表达,以及利用链霉素作为牵张激活通道(SACs)阻断剂研究SACs在其中的可能作用。结果与不牵张组HIFlα和VEGFmRNA的无表达比较,牵张可以明显增加HIFlα和VEGFmRNA的表达(P<0.05);而链霉素可以明显减少HIFlα和VEGFmRNA的表达(P<0.05);但是链霉素不能完全抑制急性牵张刺激激活的HIFlα和VEGFmRNA水平升高(P<0.05)。结论链霉素对膨胀左室致HIF1α、VEGF表达有明显抑制作用,提示心室膨胀经SACs激活胞内信号诱导HIF1α、VEGF表达。同时链霉素并不能完全抑制HIF1α、VEGF表达,暗示膨胀心室致HIF1α、VEGF表达进而引起心室肥厚尚有其他传导途径,仍需进一步研究。Objective:To investigate the effect of stretch on hypoxia-inducible factor-1α (HIF-1α) and vascular endothelial growth factor (VEGF) expression in rat hearts. Method:Isolated normal adult rat hearts were perfused by Langendorff method, stretching the left ventricle for 30 minutes by dilating the balloon. Measured the ventricular HIF-1α、VEGF mRNA expression by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) and normalized to β-actin,and analyze whether SACs play an important role in HIF-1α、VEGF expression with streptomycin (a stretch-activated channel inhibitor).Result:Compared with no stretch group, the expression of the HIF-1α、VEGF mRNA increases significantly in stretch without streptomycin group ( P < 0.05 ). Streptomycin can decrease the expression of HIF-1α、VEGF mRNA ( P < 0.05 ) , but the expression of HIF-1α、VEGF cannot be completely eliminated ( P < 0.05 ). Conclusion:Streptomycin can inhibit left ventricular dilation-induced HIF-1α,VEGF expression, which implies ventricular dilation can lead to HIF-1α,VEGF expression through SACs. But streptomycin cannot inhibit HIF-1α,VEGF expression completely, which means there are still other unknown ways to induce HIF-1α、VEGF expression and ventricular hypertrophy.
