摘 要:目的通过比较不同干预措施对气道局部炎症反应和主要免疫系统中CD4+Th2细胞活性的影响,旨在了解外源性抗IL-9抗体对哮喘潜在的治疗价值。方法40只BALB/c小鼠随机分为阳性对照组、阴性对照组、抗IL-9抗体组和地塞米松(Dex)组,建立小鼠哮喘模型,采取不同干预措施,比较各组支气管肺泡灌洗液(BALF)细胞计数、肺组织学检查。结果抗IL-9抗体和Dex均可抑制淋巴细胞、嗜酸性细胞和中性粒细胞等炎症细胞在肺内的过度聚集,降低BALF中Th2细胞因子;Dex降低支气管肺泡灌洗液(BALF)中Th1细胞因子水平,而抗IL-9抗体对其无影响。结论抗IL-9抗体和Dex均能明显减轻哮喘小鼠的肺部炎症,在气道建立起新的Th1/Th2平衡。与Dex相比,应用抗IL-9抗体建立的新平衡不会造成因细胞免疫功能受抑引起的一系列不良反应。ObjectiveTo investigate the therapeutic potential of interleukin-9(IL-9) blockage in asthma and determine the influences of anti-IL-9 antibody on airway inflammatory response and the functions of Th2-type CD4^+T cells of the mouse asthma model, we administered different interferences and inspected the various effects on asth-(matic mice.)MethodsForty BALB/c mice were devided randomly into four groups. Different interferences were taken after the mouse asthma model were established. Histological examinations were undertaken. Cell accounts and cytokines of the bronchoalveolar lavage fluid were evaluated.ResultsBoth anti-IL-9 antibody and dexamethasone could significantly depress the influx of eosinophils,lymphocytes, and neutrophils in the lung and decrease the cytokines in bronchoalveolar lavage fluid(BALF). Anti-IL-9 antibody did not change the level of Th1 cytokine in BALF, but dexamethasone did.ConclusionAnti-IL-9 antibody and dexamethasone could alleviate pulmonary inflammation of the asthma mouse, and establish a new Th1/Th2 balance in the inflammatory site. Different from dexamethasone, the new balance due to anti-IL-9 antibody would not lead to a series of effects on account of cell immunity fun-(ction) suppression in the immune response.
