“止晕栓”对家兔前庭功能障碍的药效学观察

摘　　要：研究栓剂型重碳酸钠制剂“止晕栓”的抗运动病作用，并探讨其可能的作用机理。方法应用水平旋转所致家兔眼震模型及一侧迷路破坏所致家兔自发性眼震模型，检测“止晕栓”对眼震电图各参数的影响；应用生物微球技术，检测“止晕栓”对豚鼠耳蜗血流量的影响。结果研究表明“止晕栓”直肠给药能明显抑制家兔水平旋转所致眼震的慢相速度和持续时间，作用与茶苯海明（１．６ｍｇ／ｋｇ，ｐｏ）相仿，但起效稍快，作用以给药后３０～６０ｍｉｎ最强，随剂量增加作用时间可延长，也可明显抑制一侧迷路破坏所致的自发性眼震慢相速度，尚能明显增加豚鼠耳蜗血流量。结论提示重碳酸钠栓剂直肠给药，可提高前庭兴奋阈、抑制前庭兴奋，为临床运动病、美尼尔氏病、耳源性眩晕等前庭疾患的治疗提供了一种安全、有效、方便的方法。IM To study the effect of “Zhi Yun Shuan”,a sodium bicarbonate suppository,on motion sick-ness and to probe its possible mechanisrn,so as to provide theorectical basis for the development of new antimotion sickness drugs. METHODS Nystagmus induced by horizontal rotation and spontaneous nystagmus caused by one- side labyrinth damage in rabbits were used to determine the effect of “Zhi Yun Shuan” with parameters of electronystagmogram 15,30,45,60,90 and 120 min after given the suppos-tory. Biomicroball(stained toad red blood cell)technique was used to determine the effect of“Zhi Yun Shuan” on cochlea blood flow in guinea- pigs 60 min(time of top action) after its administration,RE-SULTS“Zhi Yun Shuan”(0.3g/kg and 0.15g /kg)produced a marked inhibitory effect on slow-phase velocity and duration of nystagmus induced by horizontal rotation.The effect of this suppository is similar to that of theohydramin (1.6mg/ kg po),but its action onset is quicker with a top action at 30 and 60 min after its use.It has also an obvious inhibitory action on slow- phase velocity of nystagmuscaused by one- side labyrinth damage in rabbits. “ Zhi Yun Shuan”(0.15g/kg and 0.3 g/kg)can markedly increase cochlea blood flow in guinea-pigs 60 min after its use. CONCLUSION The resultssuggest that sodium bicarbonate suppository could increase the vestibular excitement threshold, inhibitthe vestibular excitement,thus playing a therapeutic effect.The rectal use of this drug provides a safe,effective and simple way to treat some kinds of vestibular diseases, such as motion sickness,Meniere’s syndrome and vertigo.
