肾综合征出血热病毒感染人体上皮细胞和血管内皮细胞的原位分子杂交

摘　　要：为研究肾综合征出血热（ＨＦＲｓ）患者体内病毒感染，作者应用原位分子杂交技术检测了陕西、上海、江西、沈阳和广州等地共３９例尸检病例组织中病毒ＲＮＡ的定位和分布。病毒ＲＮＡ主要定位于细胞胞浆，呈颗粒状或全浆阳性，分布于体内１６种所检测的脏器中，主要脏器中病毒ＲＮＡ的阳性检出率由高到低依次为肝（９１．６７％）、肾（８５．２９％）、肺（８４．２１％）、胃（８２．３５％）。肠（７３．６８％）、心（７０％）、脾（６１．２９％）、睾丸（５８．３３％）、脑（５６．５２％）、肾上腺（５６．５２％）和胰腺（４８．２８％）。结果显示：ＨＦＲＳ病毒具有泛嗜性感染的特点，对上皮性细胞或血管内皮细胞易感；病毒感染的部位不同，可能与不同地区ＨＦＲＳ病例所感染的病毒的血清型或毒株不同有关，由此可能导致发病机制的差异和病情轻重的不同。In order to study the target cells of hemorrhagic fever with renal Syndrome virus(HFRSV)in the patients succumbed to HFRS, in situ hybridization detection of loca-lization and distribution of virus RNA was performed on the autopsy tissues of thirty-nine patients died of HFRS collected separately from various endemic areas,including Shanxi, shenyang, Shanghai, Jiangxi and Guangzhou. HFRSV RNA was mainly locali-zed in the eytoplasm, granular or diffuse in appearance, and was widely distributed in 16 detected organs. The main organs’ positive rate for virus RNA were in the follo- wing order of frequency: liver(91.7%), kidney(85.3%), lung(84.2%), stomach(82.4%), intestine(73.7%), heart(70%), spleen (61.3%),testis(58.3%),brain(56.5%), adrenal gland(56.5%)and pancreas (48.3%).Each of these cases had at least two or more organs positive for virus RNA. Furthermore, virus RNA was also distributed in the extracellular spaces and edema fluids. Our results indicated that HFRSvirus infection was pantropic with predilection for epithelial and endothelial cells inthe body。 The difference of susceptible cells to HFRSV in cases from different endemicfoci may be related to the diversity of virus strains or serotype prevailed in differentareas and may result in variation Of HFRS in pathogenesis and severity。
