摘 要:目的:研究YAG倍频激光微束对中华大蟾蜍尾芽胚双向电泳蛋白质谱的影响。方法:以中华大蟾蜍为实验对象,以不同能量密度激光照射尾芽胚,0.5h后制备样品进行双向电泳,电泳胶扫描成像后用PDQuest软件进行分析。结果:激光照射后尾芽胚总蛋白质点数皆比对照组少,100和300μJ/cm2能量密度组总蛋白质点数减少较明显;随着照射能量的增加,新增点数逐渐增多;600μJ/cm2能量密度组消失点数较其他组少,二倍上升点数较其他组多。蛋白质点的分布及密度变化集中在分子量大于20.1kD和等电点大于4.80范围。结论:激光照射对尾芽胚双向电泳蛋白质谱有明显影响,蛋白质点的变化与激光能量密度相关。Objective:YAG double-frequency laser microbeam was applied to study the effects on protein pattern of dimensional electrophoresis of tail bud embryos of Bufo bufo gargarizans. Methods:Tail bud embryos of Bufo bufo gargarizans were irradiated with different energy density.The protein patterns were investigated by use of dimensional electrophoresis and analyzed by PDQuest. Results: Total protein spots' amount reduced and some protein spots emerged in control disappeared in laser groups and some new protein spots emerged in laser groups and some protein spots' density increasing or decreasing in different degrees .The distribution and density's fluctuations of protein spots concentrated within molecular weight of 20.1kD to 97.4kD and PI of 4.80 to 7. The higher laser energy density the more new protein spots' amount. Conclusion: Protein pattern of dimensional electrophoresis of tail bud embryos raried obviously after laser irradiation and there were relationships between protein pattern and laser energy density.
