逆针关元穴对自然更年期大鼠血脂的调节作用

摘　　要：目的:探讨逆针关元穴对自然老化的更年期雌性大鼠血脂紊乱的调节作用。方法:实验于2004-03/06在北京中医药大学针灸学院针灸机理实验室完成。选取健康9.5个月龄SD雌性大鼠56只为实验动物。选择与人类更年期综合征病理变化更为近似的自然老化术制备模型。采用随机抽签的方法将实验动物分成2组,自然对照组和逆针组。自然对照组按照自然月龄分为4组,自然10,12,14,16月龄组,每组8只。各组每天只进行抓取刺激,不作针刺。逆针组在与自然组一同养至10月龄后,采用逆针大鼠关元穴,在关元穴下约2分处将针直刺2分,后斜向上完全刺入,留针20min。留针过程中让大鼠自由活动。2次/周,持续针刺8周,并于进入更年期的12,14,16月龄后分别取材。每年龄组8只。自然对照组与逆针组同批取材。血液标本均于上午8:00～11:00采集。经处理后采用比色法测定血清总胆固醇,三酰甘油,高密度脂蛋白胆固醇,低密度脂蛋白胆固醇,按操作说明书进行。组间采用单因素方差分析。结果:56只大鼠均获得检测,无缺失值。①血清总胆固醇水平:自然12,16月龄组比自然10月龄组显著上升[(3.46±0.18),(3.19±0.16),(3.01±0.28)mmol/L,P<0.05]。逆针12,16月龄组与同龄自然组比较显著下降[(2.97±0.19),(3.46±0.18)mmol/L,P<0.05];[(2.93±0.10),(3.09±0.16)mmol/L,P<0.05]。②三酰甘油水平:自然16月龄组与自然10月龄组比较,呈显著上升趋势[(2.17±0.38),(0.99±0.09)mmol/L,P<0.01]。逆针16月龄组与自然16月龄组比较显著下降[(2.02±0.24)(2.93±0.10)mmol/L,P<0.01]。③血清高密度脂蛋白胆固醇水平:逆针12,16月龄组比自然10月龄组显著上升[(1.33±0.17),(1.42±0.12),(1.12±0.11)mmol/L,P<0.05]。逆针12月龄组与同龄自然组比较显著上升(P<0.05)。④低密度脂蛋白胆固醇水平:自然12月龄组与自然10月龄组比较呈显著上升趋势(1.77±0.22),(1.36±0.22)mmol/L,P<0.01]。逆针12,16月AIM:To investigate the regulatory effect of preventive moxibastion at Guangyuan acupoint on the blood lipid of naturally aging female climacteric rats. METHODS: The experiment was carried out in the laboratory of acupuncture mechanism, School of Acupuncture, Beijing University of Traditional Chinese Medicine between March and June 2004. Fifty-six healthy female SD rats of 9.5 months old were selected to establish models by choosing the natural aging technique which were more similar to the pathological changes of human climacteric syndrome, and then the rats were randomly divided into natural control group and preventive moxibastion group. The rats were divided into 4 subgroups according to month age: natural 10, 12, 14 and 16-month-old groups with 8 rats in each group, they were only stimulated with seizure, but not acupunctured. After fed to 10 months old together with the rats in the natural control group, those in the preventive moxibastion group were treated with acupuncture at Guanyuan acupoint: The needle was straightly inserted at 2 cun away from Guanyuanacupoint to the depth of 2 cun, and then inclined backwards until completely inserted, the needle was kept for 20 mintues, during which the rats could move freely; They were treated twice a week for 8 continuous times, and the samples were taken at 12, 14 and 16 months after climacteric period, named as 12, 14 and 16-month-old groups with 8 rats in each group. The samples in the natural control group were taken at the same period. All the blood samples were taken between 8:00 to 11:00 in the morning; After treatment, the immunoturbidimetric technique was used according to the operating instruction to test the serum levels of total cholesterol, triglyceride, high-density lipoprotein cholesterol and low-density lipoprotein cholesterol, the univariate analysis of variance was introduced for the intergroup difference.
