摘  要:本实验以正常人髂骨松质骨剪碎成的骨粒为材料,对骨粒进行预处理后,利用组织块培养法进行细胞的原代培养,原代培养形成单层后再进行传代培养。结果显示:(1)经预处理后骨基质中仍可见骨细胞,骨小梁表面及骨髓腔中无细胞残留。(2)原代细胞具有成骨细胞的形态特征。(3)经4次传代后细胞形态无明显改变。(4)细胞具有较高的碱性磷酸酶活性,且其活性并不随传代次数的增多而发生明显变化。(5)经PTH刺激后,细胞内cAMP浓度明显升高。本结果提示所培养的细胞具有成骨细胞的某些生物学行为,且细胞可能来源于骨基质中的骨细胞。能在体外观察某一影响因素对细胞形态及增殖分化的直接效应。为正常骨细胞生理、骨病理等的研究提供了一种更客观的实验手段。Abstract In order to further understand the biology of bonecells and Osteoporosis ,the authors devised the methodof the cuIture of normal human osteoblast。Eight iliac trabecular bone specimens wereobtained from patients(male 6,female 2 )undergoingnecessary surgery。The patients had abnormal ity ofbOne metabolism as judged before operation,Beforeexplanting, the bOne pieces were treated withcollagenase。The primary cultures were initiated byusing pieces of trabecular bone as explants。Theresults showed that,after treated inadvance,only osteocytes were observed in bonematrix,no cells were found on trabecular surface,theprimary cells were homogenous in appearance and hadthe morphologic characteristics,following fourpassages,the morphology of the cells was confirmed,the cells exhibited high alkaline phophatase activity,No siginificant changes of their activities was observedfollowing four passages;when exposed to the l-34fragement of human parathyroid hormone a siginificantincrease in intercellular cAMP was seen。The study suggested that,the cells had somebiological behaviors of osteoblast,and might originatefrom the osteocytes in bone matrix, The cultures canbe used to observe the direct effect of some factors ofosteoblastic morphology and proliferation,Thetechnique provided a useful system for the study ofbOne biology and bone matabolism。
