摘  要:作者报告了以聚乙烯吡咯烷酮(PVP)作为牛骨形态发生蛋白(bBMP)和基因重组碱性人成纤维细胞生长因子(bFGF)可溶性载体,将三者的混悬液经皮注射入小鼠肌肉和兔桡骨缺损处的实验研究。经不同时间点的X线摄片,组织学检查,结果发现成骨量及骨缺损愈合程度明显高于对照组(P<0.01)。注射入小鼠肌肉后及兔桡骨缺损区,观察到与新骨伴随的血管增生。证实该两种因子的联合植入,成骨作用大于单用骨形态发生蛋白,骨成熟较早,聚乙烯吡咯烷酮是骨生长因子的有效缓释载体。多种骨生长因子经皮注射,可望成为一种治疗骨折、骨缺损新的有效方法。AbstractThe effect of a composite of bovine bonemorphogenetic protein(bBMP)and recombinant basicfibroblast growth factor(bFGF) ,mixed withpolyvinylpyrrolidone as a carrier,on the healing ofsegmental defects in the rabbit radii or inducing ectopicbone formation in mice were studied.The compositewas injected percutaneously into the thigh musclepouches of the mice or the bony defects produced in therabbit radii Radiographic and histological analysiswere performed at different time intervals。The resultsshowed that all parameters were significantly greater(P<0.001) in the animals with the composite injectedthan those without the injection。 Meanwhile,increased vascularity within and adjacent to the newbOne was seen in the composite stimulated sites l4 davsafter the injection。The results revealed strongevidence of accelerated osteogenesis on the compositestimulated side,compared to those with bBMP injectedalone,Because polyvinylpyrrolidone may providecontroled, sustained release of the incorporated activesubstance,it seems promising as a carrier of purifledinductor and growth factor.These data suggest thatpercutaneous injection of bone growth factors may be apotential effective method in the treatment of fracturesor nonunion。
