摘  要:观察了中药大黄醇提物对糖尿病大鼠肾小球内多肽生长因子的影响。结果表明:糖尿病初期虽然没有明显的肾重量的增加,但此时肾小球内多肽生长因子即有明显“上调”,以链脲佐菌素注射12~24小时后最明显。应用大黄醇提物治疗后,其多肽生长因子的表达明显“下调”,并且在治疗96小时后,肾脏重量亦明显低于非治疗组。因此,提示大黄抑制糖尿病大鼠肾脏肥大的作用与肾小球内多肽生长因子表达被“下调”有关。we studied the glomerular mRNA levels encoding for properEGF,PDGF-b chain,and TGF_β in SD rats 12,24,48,and 96hours after injection of streptozotocin. At 24─hour. mRNA levels measured by dot hybridization for proper EGF,PDGF-b chain,and TGF increased 2.8-fold (P<0.05),3.1-fold (P<0. 01),and 7.3-fold(P<0. 01)respectively; at 48-hour mRNA levels forthese growth factors peaked in the glomeruli of diabetic rats as compared with control rats. Rheum Officinale(RO) treatment par-tially ameliorated the increase of mRNA levels for properEGF,PDGF-b chain and TGF_βin the glomeruli of daibetic rats. Renal hy-pertrophy was observed at 72-hour in the diabetic rats,but not in the RO-treated rats. We concluded the abnormal production ofgrowth factors in the glomeruli was involved in the early renal hypertrophy in diabetes mellitus and the effects of RO on the suppres-sion of the renal enlargement in diabetic rats may be related to its effect of down-regulated gene expression of growth factors in theglomeruli.
