狂犬疫苗接种诱生细胞免疫的研究

摘  要:狂犬疫苗对免疫接种志愿者和免疫实验小鼠能诱导产生显著的细胞介导免疫应答。免疫接种者的外周血淋巴细胞(HpBL)在体外经刀豆球蛋白A(ConA)刺激呈很强的增殖反应,增殖指数(SI)达8.5%~15.4%(P<0.001);在HpBL的ConA刺激培养上清中可测及白细胞介素2(IL-2)活性明显升高(P<0.01)。在接种者的血清中还可测及高水平α干扰素(IFN-α)(≥8×104IU/L)。免疫试验小鼠其脾淋巴细胞在体外用不同狂犬病毒株抗原进行刺激亦呈显著的特异性增殖反应,SI达7.4%~18.3%(P<0.001)。免疫鼠脾细胞的细胞毒杀伤功能亦甚强,细胞溶解率高达81.4%。将此种致敏性鼠脾细胞转输给同基因型小鼠,可使受体小鼠获得过继免疫力,保护率为50%~70%。以上初步研究结果提示,狂犬疫苗免疫接种后使机体所获得的细胞介导免疫在预防狂犬病的发病中具有重要意义。The cell-mediated immunity after vaccination with inactived rabies vaccine in volunteers or Balb/C mice was invastigated,There was a significant increase in ConA or rabies virus antigens in-vitro stimulated lymphocyte proliferative responses (P<0.001), with high proliferating indexes (8.5-15.4 for volunteer PBL and 7.4-18.3 for mouse spleenocytes, respectively), At the same time, it induced a higher level of lymphokines such as IFN-γ in serum speciments at an average≥8×1014IU/L and IL-2 (P<0.01) in the supernatant of ConA-stimulated PBL of volunteers. The cytotoxicity of spleenocytes derived from the spleen of vaccinated mice was enhanced to a high cell-lytic percentage (81.4%). Additionally, when introvenous injection of of mouse spleen lymphocytes into the syngenic mice, an adoptive immunity was obtained,which may provide the receptor mice with lethal challengedoses against rabies virus,and the protective rates may reach up to 50%-70%. All these preliminary results suggest that the cell mediated immunity induced by the rabies vaccine immunization is essential to rabies prophylaxis.
