摘  要:利用妊娠中期水囊引产的胎儿胸腺细胞,通过直接免疫荧光染色技术,借助流式细胞计,研究了PWM(Pokeweed mitogen,美洲商陆)对胎儿胸腺细胞表型的影响。结果表明:①PWM能诱导胸腺细胞CD3分子和HLA-Ⅰ类抗原表达增加,CD1分子表达减少,CD38分子变化不大。②HLA-Ⅰ类抗原和CD1分子复合表达的变化表明,胸腺细胞在PWM作用下先上调HLA-Ⅰ类抗原,表达了HLA-Ⅰ类抗原的细胞再下调CD1分子。③胸腺细胞在PWM作用下,CD4^+SP细胞和CD4^-CD8^-DN细胞逐渐增加,CD4^+CD8^+DP细胞减少。We studied the effect of PWM (Pokeweed mitogen) on the phenotypes of human fetal thymocytes. The results in dicated that PWM could induce the maturation of thymocytes by increasing the expression of CD3 and HLA class I molecules decreasing the expression of CDl ,and two-color analysis showed that the up-modulation of HLA-I was before the down-modulation of CDl. Furthermore,two-color analysis of the PWM-treated thymocytes with CD4 and CD8 showed that the percentage of CD4+CD8- and CD4-CD8- cells increased ,whereas the percentage of CD4 + CD8+ cells decreased. These results are of significance for understanding the differentiation of human thymocytes.
