摘  要:抗胸腺细胞球蛋白作为一种免疫抑制剂在预防器官和多植的排斥反应和治疗严重的再生障碍性贫血中有确切的作用。本工作在体外研究了抗胸腺细胞球蛋白(ATG)/PHA刺激人PBMC的增殖和释放细胞因子。结果为(1)^3H-胸腺嘧啶掺入试验表明ATG与PHA一样具有强的促有丝分裂活性。(2)ATH刺激的T细胞是以辅助T细胞为主(CD4/CD8〉1),在同样的条件下PHA刺激T细胞CD4/CD8的比值小1。(3)Anti-Thymoglobulin (ATG) has been used as an immunosuppressive agent to prevent transplant rejection and to treat a number of severe aplastic anemia. We have studied that ATG/phytohemagglutinin (PHA) stimulated peripheral blood mononuclear cells (PBMC) to proliferate and release cytokines in vitro. In conclusion; (1) ATG/PHA had their mitogenic effects on PBMC in vitro 3 H-thymidine incorporation. (2) ATG stimulated the helper-inducer cells (CD4/CD8>1) in PBMC. This ratio was different from that observed in short-term culture of T cells stimulated with PHA under the same conditoins (CD4/CD8<1). (3)Supernatants of ATG-primed T cells had a promoting effect on granulocyte-macrophage colony forming unit (CFU-GM) growth. On the contrary, IL-2, TNF-a and IFN-r levels were higher in PHA primed T cells supernatants,producing significant inhibition of CFU-GM growth. The results showed that T cells became activated and entered into a proliferative phase upon ATG treatment. Since activated T cells release cytokines with regulatory functions on the immune response and on hematopoiesis.it is possible that some of the therapeutic effects of ATG may be related to an immunoregulatory fuction rather than imunosuppressive function.
