摘 要:采用DynalM-450CD_(34)免疫磁珠分离纯化脐血CD_(34) ̄+细胞,并探讨了不同细胞因子[干细胞因子(SCF)、白细胞介素6(IL-6)、白细胞介素3(IL-3)及红细胞生成素(Epo)]组合对脐血CD_(34) ̄+细胞的体外培养和扩增作用。结果表明:①经DynalM-450CD_(34)免疫磁珠分离的脐血CD_(34) ̄+细胞,其纯度达85%~。95%;②甲基纤维素培养体系中,在不同组合的细胞因子作用下,脐血CD_(34) ̄+细胞的集落产率明显不同,单用IL-6最低,SCF、IL-6、IL-3及Epo联用产率最高;③在低氧液体培养体系中,IL-6和IL6+Epo不能有效地扩增脐血CD_(34) ̄+细胞,而其它细胞因子组合均可产生明显的扩增作用;其中加Epo的组合扩增效果显著高于无Epo的相应组合。脐血CD_(34) ̄+细胞分离、纯化及其体外扩增的研究对于脐血造血细胞库的建立,脐血造血细胞移植和基因导入的研究均具有重要意义。D_(34) ̄+ Cells from umbilical cord blood were purified by Dynalbeads M450 CD,. immunoselection system and cultured in the presence of various combina-tions of cytokines , including stem cell factor (SCF) .interleukin-3 (IL-3 ) , IL-6 and erythropoietin (Epo ).The results showed that OI 85%~ 95/00 of the cellsseparated by the immunobeads method were CD_(34) positive cells. ②In methylcellulose culture, the colonyyields of purified umbilical cord blood CD_(34) ̄+ cells weredifferently stimulated by various cytokine combina-tions. IL-6 alone had the lowest colony yield, whilethe combination of SCF + IL-3 + IL-6 + Epo had thehighest colony yield. O3 In suspension culture, IL-6alone or IL6-Epo had little expanding effect on um-billical cord blood CD_(34) ̄+ cells, while other cytokinecombinations could ecpand umbilical cord blood CD_(34) ̄+cells at different levels. The expanding effects of thecombinations containing Epo were much superior totheir counterparts without Epo.The study of umbilical cord blood CD_(34) ̄+ cells separation and their in vitro expansion open up the possibilityof the establishment of umbilical cord blood hematopoi-etic stemlprogenitor cells bank and clinical transplan-tation and gene transduction with these cells.
