摘  要:用辣根过氧化物酶标记McAbF(ab')_2、McAbLV48A、McAb3D_8和McAbA_(35),分别对27例EHF患者尿液中融合细胞(FC)进行免疫酶染色。结果McAbF(ab')_2检出率为27/27(100%);McAbLV48A为19/27(70.37%);McAb3D_8为20/27(74.07%);McAbA_(35)为2/27(7.41%)。人胚肾细胞感染EHF病毒后,在偏酸性(pH6.0)条件下可以产生特异性细胞融合现象,并经电镜证实了细胞之间膜的融合。此结果表明EHF病毒可以导致人肾细胞融合,EHF患者尿液中FC的形成可能与EHF病毒膜蛋白在宿主细胞膜上的表达有关。Using McAb immunoperoxidase technique, 27 cases of EHF patients urine, in which the fusion cells (FC ) had been found,were detected and Hantavirus antigens were found on or in all the FC.At the same time, for localizing the structural proteins of Hantavirus (such as 48kd nucleocapsid protein NP, and two envelope glycoproteins (G1 68kd,G2 55kd ) ,LV48A(McAb direct against the G2 of A9 strain),3 D8 ( McAb against the hemagglutinin of Chen strain ) , and A35 (McAb against NP of A9 strain ) were used in detecting 27 cases of FC. The result ahowed that 19 presented positive reaction to LV48A,20 to 3D8and 2 to A35.This result indicated that the formation of the FC should be closely related to the Hantavirus glycoproteins, or hemagglutinin expressed on the membrane of host cells.After human embryo kidney cells (HEKC ) were infected by hantavirus, the cell fusion was found to occur under acidic conditions (pH6.0 ) .Under electromicroscope, the cell fusion was confirmed.
