两种不同给药途径对松针提取液抗突变效应的比较研究

摘　　要：采用腹注和灌喂两种给药方式，研究了松针提取液对环磷酰胺诱发小鼠骨髓细胞微核形成率的影响，以观察松针提取液的抗突变效应以及给药途径对抗突变效应的影响。实验结果表明，松针提取液的二种不同给药方式对环磷酰胺的致突变性均有抑制作用，使微核率显著下降，与环磷酰胺组相比，经统计学处理有极显著性差异（Ｐ＜０１），但松针提取液抗突变效应并非剂量越大，效果越佳，只有在一定的剂量范围内，才能显示出最佳抗突变效应。亦即剂量与微核率只有在一定剂量范围内才呈现显著的负相关，相关系数分别为ｒ＝０．９８４２（ｐ＜０．０５）及ｒ＝—０．９７８２（ｐ＜０．０５）。两种不同给药方式对松针提取液的抗突变效应，经ｘ２检验无显著影响（ｘ２＝４．３７０，ｘｈ＝７．８１５，ｘ２＜ｘＺ。。），实验结果还提示，松针提取液在低剂量时不呈现致突变效应，但灌喂或腹注达一定剂量时却有一定的致突变性，因此在利用松针提取液进行肿瘤防治时，选择适当剂量至关重要。The antimutagenic effect of pine needle on frequency of micronucleated polychromatic erythrocytes (MNPCEs) in mouse bone marrow induced by cyclophosphamide (C.P. ) was studied. The results showed that the pine needle extract can inhibit the mutapenic toxicity following both the intraperitoneal injection and the oral gavage. The differences compared with the positive control were statistically significant (P<0.01). However, the optimal antimutagenic effect could be observed only in a suidtable range. Our results showed that a clear negative dose-effect relationship could be observed in a certain range of doses between 500 mg/kg body weight and 2000 mg/kg body weight following two routes of administration (r = -- 0. 8942, -- 0. 9782, respectively). There were no statistically significant differences between the effect following the two routes of administration. Our research also showed that the pine needle extract could exhibit some mutagenic effect at high dose (4000 mg/kg body weigh ), and the pine needle extract could suppress the mutagenic activity of cyclophospha-mide. The micronucleus frequency and frequency of SCEs in the mouse bone marrow were reduced significantly and indicated the distinct negative correlation with the dose of pine needle extract. So the choice of proper doses is important when considering its practital value.
