人核糖体磷酸蛋白P2基因的克隆及鉴定

摘　　要：利用聚合酶链式反应技术（ＰＣＲ），从人肺巨细胞癌ＰＬＡ－８０１母系细胞系统克隆化高转移细胞株Ｄ中特异性扩增Ｐ２ｃＤＮＡ（ＨｕｍａｎｒｉｂｏｓｏｍａｌｐｈｏｓｐｈｏｐｒｏｔｅｉｎＰ２ｇｅｎｅｃＤＮＡ），并将扩增产物克隆入ｐＧＥＭ－３Ｚ载体，获得了重组质粒则ｐＭＰ２，对其中两个克隆ｐＭＰ２－１、ｐＭＰ２－２的Ｐ２ｃＤＮＡ进行了序列分析。结果表明：ｐＭＰ２－１的Ｐ２ｃＤＮＡ序列与文献报道基本一致，只是在第１４８位碱基处发生了置换，由Ｃ→Ｇ，而ｐＭＰ２－２的Ｐ２ｃＤＮＡ在１４８位碱基未发生改变，但在第１１０位碱基却发生了缺失，在第２００位碱基由Ｇ→Ｃ。P2 gene cDNA was specifically amplified by PCR technique from strainD with high metastastic potential of human lung giant cell carcinoma cell line,( PLA-801)and then cloned into the polylinker site of vector pGEM-3Z. Two recombinantclones(pMP2-1 and pMP2-2 ) were sequenced.The sequencing data revealed that the P2cDNA of recombinant clones is almost the same as the one reportedbefore,In pMP2-1,C was substituted by G at position 148,in pMP2-2, G was replaced by C at position200,and an absence of A was found at positlon 110.
