摘  要:本实验观察川芎嗪对家兔实验性膜性肾炎时血小板功能变化的影响,并探讨血小板功能异常与膜性肾炎病理变化的关系。结果表明:川芎嗪对本型肾炎的发展有较明显的治疗作用。其作用机制可能是通过降低血小板聚集性,抑制血小板TXA_(2)生成和活性,增加血小板cAMP含量,抑制血小板内容物的释放,调节血浆和肾组织TXA_(2)/PGI_(2)的平衡,从而稳定血小板而减轻肾小球基底膜的损伤,阻抑渗出,减轻蛋白尿,从而改善肾功能,减轻肾组织的病理损伤。Effect of Tetramethylpyrazine(TMP)on platelet function in Japanese wh-ite rabbits with membranous glomerulonephritis (MGN)was studied to find out the re-lationship between abnormal platelet function and pathomorphological changes of MGN.The results showed that TMP iuhibited platelet aggregation, and TXA_2 synthesis and itsactivity in platelet.TMP also increased synthesis of PGI_2, enhanced cAMP contents inplatelet, and inhibited the release of 5-hydroxytryptamine from platelet. At the same tirne,the drug reduced the amount of proteinuria, inhibited the fibr in from depositing alongthe glomerular basement membrane and Bowmen′s capsule.The pathomorphological lesionwas very much mitigated in the therapeutic group compared with MGN group. It sug-gests that the effect of TMP on stabilizing platelet may be an important factor in allevi-ating pathomorphological lesion and inhibiting the development of MGN.
