用双池培养研究颗粒细胞与内泡膜细胞的旁分泌调节作用

摘　　要：采用双池培养系统研究颗粒细胞（Ｇ－Ｃ）与内泡膜细胞（Ｔ－Ｃ）在甾体激素生成中的旁分泌调节作用。实验选用小母猪的卵巢组织，分离出Ｇ－Ｃ和Ｔ－Ｃ，巳分别进行单独培养（单培）或双池培养（双培），内地为Ｇ－Ｃ，外池为Ｔ－Ｃ。４８小时后用放射免疫法（ＲＩＡ）测定培养液中雌二醇（Ｅ２）、睾酮（Ｔ）及孕酮（Ｐ）含量。结果显示：（１）在单培时Ｇ－Ｃ产生极少量Ｅ２，当其与Ｔ－Ｃ双培时则分泌大量Ｅ２；Ｐ和Ｔ产量在双培时比单培时均有增加。（２）Ｔ－Ｃ在单培时产生大量Ｔ，当其与Ｇ－Ｃ双培时，在培液中可测出少量Ｔ；Ｐ和Ｅ２产量在双培时比在单培时均有明显下降。结果表明：在甾体激素生成过程中，Ｇ－Ｃ和Ｔ－Ｃ之间呈现一种旁分泌的调节作用。Da Lian Family Planning Research Institute Objectives :To investigate the paracrine interaction in steroidogenesis between granulosa cells and thecal cells. Design :An in vitro culture study. setting :Endocrinology laboratory in National Research Institute for Family Planning Methods :Granulosa cells and thecal cells were separated from the porcine ovarian follicles and were cul-tured in McCoy’s 5a medium in a dual chamber system with Matrigel. Granulosa cells were placed in the inner chamber,while the thecal cells,in the outer chamber. Estradiol(E2),progesterone (P)and testos-terone (T )in the medium were measured with RIA after 48hrs incubation. Results :In single culture of granulosa cells very small amount of E2 Was produced ,but when co-cul-tured with thecal cells,the amount of E2 and P was increased.Thecal cells produced large amount of T insingle culture, but in very small amount when co-cultured with granulosa cells;likewise,the amount of E2 and P decreased. Conclusions :Granulosa cells produced estrogens only in the presence of an androgen substrate when cul-tured singly. In a coculture system the granulosa cells in the inner chamber can produce estrogens throughthe aromatization of testosterone which prOduced by the thecal cells in the outer chamber.Thus the dualchamber system displayed a paracrine regulatory interaction between the two cells in steroidogenensis.
