用PCR技术产前诊断人巨细胞病毒感染

摘　　要：用ＥＬＩＳＡ法筛选早孕妇女血人巨细胞病毒ＩｇＭ（ＨＣＭＶ－ＩｇＭ），在孕１２～２０周用聚合酶链式反应（ＰＣＲ）技术检测其羊水中ＨＣＭＶ－ＤＮＡ，对胎儿巨细胞病毒感染作出产前诊断。在３８４例孕妇中，筛选出血ＨＣＭＶ－ＩｇＭ阳性２６例为实验组，其羊水ＨＣＭＶ－ＤＮＡ检出率为３４．６２％；从２５８例ＨＣＭＶ－ＩｇＭ阴性中选出２０例为对照组，其羊水ＨＣＭＶ－ＤＮＡ检出率为１０．０％，两组比较有显著性差异（Ｐ＜０．０５）．对两组中共８例进一步查新生儿脐血ＤＮＡＰＣＲ均为阳性，证实有ＨＣＭＶ感染。Human Cytomegalovirus IgM (HCMV-IgM)in blood of 384 pregnant women in 12-20 weeks of gestation was detected by ELISA, and 26 pregnant women of them were positive for HCMV-IgM in blood. The amniotic fluid of these 26 cases(as study group) and 20 negative cases(selected as controls) was collected and HCMV-DNA was detected by polymerase chain reaction (PCR) to diagnosis fetal HCMV infection before birth. The results showed that 9 of the 26 cases and 2 of the 20 were positive for HCMV-DNA, constituting a total positive rate of 23.91% (11/46). Umbilical cord blood was detected by PCR in 11 positive cases;all of them were positive for HCMV-DNA PCR.
