鼠抗人A_1亚型红细胞单克隆抗体研制及其免疫学性质研究

摘　　要：应用杂交瘤技术，以２种抗原分别免疫ＢＡＬＢ／Ｃ小鼠。从３次细胞融合中，筛选出４株分泌高滴度、高特异性抗Ａ１血型物质单克隆抗体的杂交瘤细胞系。经多种免疫化学试验证明：其中Ｂ５２３－Ｈ１０、Ｄ２４３－Ｈ９２株杂交瘤扩大培养上清液仅凝集Ａ１红细胞，而不凝集Ａ２、Ｂ、Ｏ型红血胞。它们结合部位的结构互补于：人工接上脂肪链的Ａ－活性三糖。从而表明该２株单抗为首次获得的抗Ａ１亚型红细胞单克隆抗体。不仅为研究Ａ１亚型血本质提供了极好的材料，也为医学临床提供Ａ１亚型血分型试剂准备了条件。Using hybridoma technique,We prepared a group of byhridomas by different immunization procedures.They secreted high level and high specific monoclonal antibodies to A1blood substances.It was demonstrated that the tissue culture superatant of two of them(D234-H9 and B523-H10)could only agglutinate human A1-type erythrocytes specifically,but could not agglutinate A2,B and O type erythrocytes by various immunochemical assays.The complemental structure of their combining site was most complementary to the synthetic A blood group nonreducing trisaccharides glycosidically linked to -O(CH2)8 COOH.The results shaw that this is the first report of two monoclonal antibodies to human A1-type erythrocytes which will offer excellent material for the study of the nature of A,subgroup blood.They can also offer the grouping reagents for A1 subgroup blood in clinical medicine.
