摘 要:gp130结合分子(GAM)是以gp130胞浆内段为探针新近克隆成功的一个分子,其功能仍不清楚。为探讨GAM在gp130信号传导中的可能作用,我们将不同长度的gp130变异体转染Hep3B细胞,观察GAM与gp130的免疫共沉淀。结果表明GAM可与gp130胞浆内近胞膜部分相结合。当GAM、gp130和JAK激酶共转染COS细胞时,GAM的表达可明显抑制JAK激酶对gp130的磷酸化。以上结果提示,GAM可能作为一个抑制分子参与gp130的信号传导。Abstract gp130 associated molecule(GAM) is a newly cloned molecule by using cytoplasmic part of gp130 as probe.To investigate its possible role in gp130 signal transduction,we trans feeted different length Of gp130 mutant cDNA into Hep3B cell,and checked the co-immunoprecipitation of GAM and gp130.The result indicated that GAM associated with gp130 at its membrane proximal region.When the cDNA of GAM,JAK kinase(JAK1,JAK2 and TAK2) and gp130 were co-trans fected into COS cell,the expression of GAM inhibited both the association of JAK kinase with gp130 and the tyrosine phosphoralation of gp130 by JAK kinase.The result suggest that GAM may fountion as a inhibiting molecule in the signal transduction of gp130.
