毛细管GC法同时测定咪唑安定及其代谢物的血药浓度

摘  要:用气相色谱法测定了人体血浆中咪唑安定及其代谢物的含量。以Ro21-2212作内标,在弱碱性条件下用氯代丁烷-二氯甲烷(7∶3)提取样品,以氮气为载气,用HP-5大口径毛细管柱和氮磷检测器进行分析。结果咪唑安定、4-羟基和α-羟基咪唑安定平均回收率分别为99.62%、99.55%和98.86%,最低检测限分别为1、5和5ng/ml,日内和日间变异系数均小于8%。提示该法灵敏度高,专一性强,且内源性物质和同时使用的药物均不干扰。用本法还测定了全麻择期手术患者的药代动力学参数。A GC method is deseribed for measuring midazolam and its metabolites,α-hydroxy and 4-hydroxy midazolam simultaneously in human plasma.Compounds were extracted with chlorobutane-dichloromethane (7:3) at basic PH with Ro 21-2212 added as internal standard. A portion (2 μl) was analysed with N2 as carrier gas on a wide-bore capillary column of HP-5 and N-P detector. Mean recovery was 99. 62% for midazolam, 99. 55% for 4-hydroxy midazolam, 98. 86% for α-hydroxy midazolam and the detector limit was 1, 5. 5 ng/ml, respectively. The coefficients of variation for the inter- and intra-day were all less than 8%.No interference was found from endogenous compounds, or other commonly used drugs.Those suggest that this method can be applied to therapeutic drug monitoring and pharmacokinetic studies.
