摘  要:作者采用诱导剂和杂交瘤细胞同步生产单克隆抗体(McAb)腹水法,研究比较了不同成分诱导剂生产McAb腹水的效果.结果表明以液体石蜡与不完全佐剂的等量混合剂(LPIFA)诱导McAb腹水产生的效果优于以不完全佐剂(IFA)或降植烷(pristane)为诱导剂生产腹水的效果.LPIFA法是IFA或降植烷法腹水产量的1.4~2.2倍、McAb产量的1.6~2.5倍;每鼠平均诱生腹水约57~76ml,其中单只最高一次达110ml.并且应用LPIFA同步转种杂交瘤细胞生产腹水法有省时、省工的优点.The effects of rapid induced rat ascites formation and increased monoclonal antibody(McAb)anti- HFRS virus production by different inducers were studied.Incomplete Freund’s adjuvant(IFA ),pristane orv/v mixture of paraffin liquid and incomplete Freund’s adjuvant (PLIFA)were used to induce ascitic fluid in Lou/c rat. The best results were obtained following injection of 2 ml PLIFA at the moment of intraperitoneal transfer of rat hybridoma cells. Ascites production was increased by as much as 1.4~2.2 times and yield of McAb by more than 2 times compared with pristane or IFA treated rats.The mean ascites production was 57 ~76 ml per rat by PLIFA,and the highest was 110 ml.Induction of PLIFA may also save time and decrease labour intensiveness.
