摘  要:为了观察血小板激活因子(PAF)在急性低压缺氧中的作用,选用了天然PAF拮抗剂Taponin进行缺氧干预实验.结果发现,用0.5g/kg剂量给大鼠灌胃给药8次,对急性低压缺氧引起的大鼠心肌、肝功能和内皮细胞损害有较好保护作用,血液中的CPK、LDH、α-HBD、AST、ALT、γ- GT以及 ET含量均未见明显升高,和对照组相比,差异无显著性(P>0.05).另外,本实验还发现,20%的低浓度乙醇对急性低压缺氧也有一定的保护作用.Acute hypobaric hypoxia may produce many kinds of effects on the organism. We have found in early investigations that the amount of circulatory endothelial cells was increased, plasma en-dothelin content was greatly increased, and that ultrastructure of the myocardium, activities of myocar-dial enzymes and hepatic functions changed greatly after acute hypobaric hypoxia. All these facts suggested that there were important effects of acute hypobaric hypoxia on myocardium, liver and vascular endothelial cells. It was found that hypoxic state may cause significant production and release of platelet activating factor (PAF) in platelets, white cells and endothelial cells, which may result in damage of the various organs. In order to investigate the effect of PAF during acute hypobaric hypoxia, Taponin - a kind of natural anti - PAF substance was used in the experiment to observe its protection against the harmful efffects of acute hypobaric hypoxia. The results showed that gastric irrigation with Taponin 0.5 g/kg for eight times had certain protective effects against the damage of myocardium, hepatic functions and endothelial cells caused by acute hypobaric hypoxia. The contents of CPK, LDH, α - HBD, AST, ALT, γ - GT and ET after hypoxia were not significantly different from those in the control. This means that there is certain relationship between PAF and the harmful effects of a-cute hypobaric hypoxia.
