摘  要:本实验采用全胚胎培养(WEC)技术研究了氟对体外培养48小时大鼠胚胎的发育毒性作用。结果表明:氟浓度为5μg/ml时,仅对脏层卵黄囊(VYS)血管形成产生抑制作用;10μg/ml时,反映胚胎生长发育和形态分化的各项指标均明显低于对照组,畸胎率为55.6%;20μg/ml时,培养胚胎无一正常,死胎率达66.7%;电镜观察可见VYS上皮细胞表面的微绒毛变短、脱落等形态改变,溶酶体、线粒体等细胞器的结构也发生改变,提示VYS的病理改变可能与氟的致畸机制有关。We tested the toxicological effects of fluoride on rat embryo development for 48h with whole embryo culture technique in vitro.The results showed the blood circulation of visceral yolk sac vessel formation was only inhibited with exposure to 5.0μg/ml fluoride.All parameters connected with embryonic growth,development and morphological differentiation were smaller than that of control group.In 10.0μg/ml,group,the incidence of abnormal embryo was 55.6%.At fluoride concentration 20. 0μg/ml,the embryonic mortality was up to 66.7%.Using electron microscopy to examinate the embryonic visceral yolk sac showed the changes of ultrastructures of microvilli、mitochondria、lysosomes、endoplasmic reticula et al.The pathological changes of visceral yolk sac structure would be related to teratogenic mechanism of fluoride.
