乙酰胆碱和5—羟色胺对日本血吸虫雄性成虫细胞电活动的影响

摘  要:微电极细胞内记录技术应用于研究乙酰胆碱(Ach)和5—羟色胺(5—HT)对日本血吸虫雄性成虫皮层和肌细胞电活动的作用。实验结果显示:(1)Ach对虫体机械活动和肌细胞(n=96)电活动有明显抑制作用。阿托品能拮抗(Acm)的作用,而使虫体活动和肌细胞电活动增强;(2)5—HT对虫体皮层细胞(n=123)和肌细胞(n=107)静息电位有去极化作用,去极化幅值为2—6mV,并能明显增强细胞放电频率。同时,也增强虫体活动度。赛庚啶能拮抗5—HT对虫体的作用;(3)虫体机械收缩活动与肌细胞放电频率成正比的关系。虫体细胞对Ach和5—HT的作用具有明显量效反应。5—HT的作用与细胞膜Ca2+的流通有关。Actions of acetylcholine(Ach)and 5-hydroxtrypamine (5-HT)on electrical activity of tegumental and muscle cells in Schistosoma japonicum were studied by the intractllular recording techniques with microelectrode. The results shown that(1)Ach applied to male adult Sjaponicum preparation caused significant inhibition on mechanical contraction of the worm and electrical activity of muscle cells.Atropine might antagonize the effects of Ach;(2)5-HT in perfusing solution made resting potentials of tegumental and musele cells showed depolarizing responses,depolarizing amplitude was 2 to 6 mV,and it increased apparently discharge frequenee of muscle eells in the worm.At the same time,5-HT also strengthened motor activity of the parasite.The effects of 5-HT might be antagonized by cyprohetadine in the preparation;(3) There was a direct ratio relationship between mechanical contractive activity of the S.japonicum and discharge frequence of muscle cells.Furthermore,these responsed had obvious dose-effects characters to Ach and 5-HT.The effects of 5-HT might be correlated with calcium afllux on cellular membrane in male adult S.japonicum..
