摘  要:中药海蒿子干粉经氯仿脱脂、热水抽提、乙醇沉淀和DEAE-SephadexA-25柱层析分离,得到多糖SPⅠ、SPⅡ和SPⅢ,总糖含量分别为52.40%、51.60%、38.80%。用醋酸纤维薄膜电泳鉴定SPⅢ级分含有两种组分,SPⅠ和SPⅡ级分均为单一组分。紫外光谱对三种级分多糖溶液在220~400nm范围扫描,显示在260nm、290nm处SPⅠ、SPⅡ级分无核酸和蛋白吸收峰。红外吸收光谱表明三种级分多糖结构中均含β构型吡喃糖苷键。经酸降解、薄层层析和气相色谱-质谱分析,证明其单糖组成均为木糖、岩藻糖、半乳糖、果糖、甘露糖、葡萄糖,其中岩藻糖含量最高。The Polysaccharide SP Ⅰ ,SP Ⅱ and SPⅢ components were isolated from the sargassum Pallidium powder by Chloroform defatting, boiling water - extracting, ethanol - precipitation and DEAE - Sephadex A - 25 column chromatography. The content of total sugar were 52.40%, 51.60% and 38.80%. The SPⅢ showed two compoents in cellulose acetate membrane electrophoresis, while the SP Ⅰ and SP Ⅱcomponents showed single component. There were no nucleic acid and protein absorbance at 260nm and 290nm on the ultraviolet spectrum of SP Ⅰ and SP Ⅱ. The data of IR revealed that SP Ⅰ , SP Ⅱ and SP Ⅲ were chiefly linked by β- glycosidic bonds. The chemical composition analyzed by TLC and GC - MS showed that they were composed of xylose, fucoidan, galactose, fructose, mannose and the glucoses; and the fucoidan was primary component.
