摘 要:目的测定桑树资源中1-脱氧野尻霉素(DN J)并分析其生物活性。方法以氯甲酸芴甲酯(FM OC-C l)柱前衍生RP-HPLC荧光检测法对DN J进行分析;在酶水平测定样品的α-糖苷酶抑制曲线,在离体水平用反转肠囊实验测定样品的α-糖苷酶抑制活性。结果建立了DN J的定量方法,并应用于桑树资源中DN J的定量;这些中药材与DN J对照品呈现了平行的α-糖苷酶抑制曲线,对淀粉水解和葡萄糖吸收具有较好的抑制作用。结论桑树资源可以作为α-糖苷酶抑制剂治疗糖尿病。Objective To determine 1-deoxynojirimycin (DNJ) from mulberry resources and analyse their bioactivities. Methods Determination of DNJ by derivatization with 9-fluorenylmethyl chloroformate (FMOC-C1) and followed by reversed-phase high-performance liquid chromatography (RP-HPLC) and a fluorescence detector; the inhibitory curves on α-glucosidase at the enzymatic level and the activities to α-glucosidase in vitro were observed in the everted sac experiment. Results A reliable quantitative method suitable for assays of DNJ from mulberry resources has been developed. The inhibitory curves of the extracts on α-glucosidase were parallel to those of DNJ. The extracts can inhibit hydrolyzation of starch and absorption of glucose. Conclusion The medicinal parts from mulberry resources can be used for diabetic therapy as α-glucosidase inhibitor.
