摘　　要：目的:建立测定血清中更昔洛韦的RP-HPLC法。方法:以阿昔洛韦为内标,采用C18柱及保护柱,流动相为甲醇-水 (60:40),磷酸调pH6．5,流速1．0 ml·min-1,紫外检测波长254 nm。结果:更昔洛韦与内标分离良好,更昔洛韦保留时间9．25 min,阿昔洛韦保留时间为11．23 min。回归方程为A=107．9C-0．202(r=0．999 7)。其线性范围为0．05-10μg·ml-1。结论: 该法操作简便,测定结果精密度高,重现性理想,适用于药动学研究。Objective： To set up RP-HPLC assay to determine the content of ganciclovir in human serum. Method. Aciclovir was used as internal standard. Analysis was carried out on a chromatographic system using C18 column and UV detection at 254 nm. Methanol-water （60： 40） was used as mobile phase, adjusted pH 6.5 with phosphoric acid. Velocity of flow was 1. 0ml·min^-1. Result： The Ganciclovir and internal standard departed well. The retention time of Ganciclovir and aciclovir was 9.25 min and 11.23 rain respactively. The regress equation was as follows： A =107.9C -0.202 （r =0.9997）. Calibration was linear from 0.05 to 10 μg.ml^-1.Condusion ： This assay is easy to operate and to be repeated. The accuracy of the assay is high. It appears to be suitable for the pharmacokinetic studies.
