摘  要:目的开展血吸虫表达基因谱的研究,寻找新的疫苗候选分子和药物靶标。方法应用表达序列标签(EST)和电子克隆策略。结果获得了552个EST序列和487个电子延伸序列,其中104个EST序列在延伸前未表现同源性,而延伸后表现出有意义的同源性;获得了日本血吸虫基因表达谱的信息以及发现了有潜在药物和疫苗价值的新基因序列。结论本研究为日本血吸虫基因表达谱的研究提供更有效的研究思路。Objective To study the gene expression profile of Schistosoma japonicum (S.japonicum) and identify novel genes, for to the discovery of potential targets for drugs and vaccines to combat schistosomiasis. Methods We selected single recombined clones randomly from cDNA library of S. japonicum adult worm and got the sequences, generated expressed sequence tags (ESTs), and then ESTs were elongated by the platform of in silico elongation. Results In the study, 552 ESTs and 487 elongated sequences were obtained. Sixteen sequences after elongation showed high homology with the sequences from Genbank database. One hundred and four elongated sequences have significant homology but no homology at all before elongation. Novel genes and the expression profile of S. japonicum were identified. Conclusion Analysis of the torrent sequence information using ESTs and in silico elongation leads to rapid identification of the novel genes and the expression profile of S. japonicum. In particular, there would support the discovery of potential targets for drugs and vaccines to combat schistosomiasis.
