辅酶A口服剂与泛硫乙胺对家兔食饵性高脂血症作用的比较

摘　　要：目的:比较辅酶A口服剂(辅酶A+辅酶A保护剂)与泛硫乙胺口服给药对家兔食饵性高脂血症的作用。方法:56只雄性新西兰白兔随机分为7组,每组8只。其中1组饲以普通饲料作为正常对照组,另外6组饲以高脂饮食20d制备高脂血症模型,20d后随机选择3组每日给予辅酶A保护剂[825mg/(kg·d)]+高中低3个剂量的辅酶A[100、800、1500U/(kg·d)]。其余3组分别给予辅酶A前体药———泛硫乙胺[1200mg/(kg·d)]、辅酶A保护剂[825mg/(kg·d)]、辅酶A原药[800U/(kg·d)],口服给药20d。治疗前后测定动物血总胆固醇、三酰甘油、低密度脂蛋白胆固醇、高密度脂蛋白胆固醇水平。结果:给药20d后,辅酶A口服剂高、中剂量组动物总胆固醇、低密度脂蛋白胆固醇显著降低,其效果与泛硫乙胺无显著差别。大剂量辅酶A可降低家兔三酰甘油水平,泛硫乙胺无此作用,辅酶A原药未见明显的降脂作用。结论:辅酶A口服剂治疗高脂血症效果可能优于泛硫乙胺。Objective：To compare the effects of an oral preparation of Coenzyme A （CoA） with pantethine on hyperlipidemia in rabbits fed on high-lipid food. Methods： Totally 56 male rabbits were equally randomized into 7 groups. Rabbits in control group were fed with normal diet, and hyperlipidemia rabbit models were established by feeding high-lipid food for 20 d.Three hyperlipidemia groups were administered intragastrically with CoA protective agent （825 mg/[ kg · d]） plus different doses of CoA （100, 800 and 1500 U/[kg· d]） after another 20 d. The other 3 groups were administered with pantethine （1 200mg/[kg· d]）, CoA protective agent （825 mg/[kg· d]） and originalCoA （800 U/[kg· d]）, respectively. Results： Oral CoA at 800 and 1 500 U/（kg· d） for 20 d reduced total cholesterol and low density lipoprotein cholesterol significantly, and there was no significant difference between the effect of oral CoA and pantethine. Oral CoA at 1 500 U/（kg · d） also reduced triglyceride while pantethine had no such effect, but original CoA had no effect on cholesterol and triglyceride. Conclusion： Oral preparation of CoA may be better than pantethine in the treatment of hyperlipidemia in rabbits.
